View Andrew Murphy's current disclosuresGd-DTPA labelled dextran is an intravascular MRI contrast agent. Dextran is a polysaccharide consisting of a polymer of glucose molecules with a molecular weight between 75,000-100,000. Dextran has a high level of safety and is broken down more rapidly than albumin. Approximately 15 Gd-DTPA molecules are attached to each dextran molecule with an easily hydrolyzable bond. It is hoped that this will reduce the long term in vivo retention seen with Gd-labelled albumin. The trade off is a shorter biological half-life of 43 minutes. In a rat model, satisfactory enhancement of liver, spleen, kidneys, myocardium, and brain is seen for up to 1 hour. The dose of dextran Gd-DTPA in these studies was 0.01-0.05 mmol/kg 1.
A variation of this contrast agent has been commercialized as Riovist (Schering) 2.
